چکیده

In mouse ovaries, the enzyme 3 beta-hydroxysteroid dehydrogenase (HSD) is distributed between microsomes and mitochondria. Throughout the follicular phase of the estrous cycle, the HSD activity in microsomes is predominant; whereas, after LH stimulation, HSD activity during the luteal phase is highest in the mitochondria. The current study examined whether or not LH stimulation always results in an increase in mitochondrial HSD activity. This was accomplished by measuring the HSD activity in microsomal and mitochondrial fractions from ovaries of pregnant mice. These animals have two peaks of LH during gestation, and one peak of LH after parturition. It was found that mitochondrial HSD activity was highest after each peak of LH. It is proposed that mitochondrial HSD is essential for the synthesis of high levels of progesterone. The increase in HSD activity in mitochondria after LH stimulation occurs because: 1) LH initiates the simultaneous synthesis of HSD and the cholesterol side-chain cleavage enzyme (P450scc); and, 2) HSD and P450scc bind together to form a complex, which becomes inserted into the inner membrane of the mitochondria. High levels of progesterone are synthesized by mitochondrial HSD because: 1) the requisite NAD+ cofactor for progesterone synthesis is provided directly by the mitochondria, rather than indirectly via the rate limiting malate-aspartate shuttle; and, 2) the end-product inhibition of P450scc by pregnenolone is eliminated because pregnenolone is converted to progesterone.

Relationship between ovarian delta 5-3 beta-hydroxysteroid dehydrogenase activity and implantation in non-lactating and lactating mongolian gerbils (Meriones unguiculatus).

The distribution and intensity of delta 5-3 beta-hydroxysteroid dehydrogenase (3 beta-HSD) activity was examined histochemically in ovaries from Mongolian gerbils during pregnancy and undergoing lactational delay. In all of the ovaries examined the corpora lutea showed the strongest level of activity, but like the interstitium and granulosa, there was no relationship between the enzyme pattern ...

Effect of Aqueous Extract of Physalis Alkek-engi Fruits on the Activity of Ovarian 3beta- and 20alpha-Hydroxysteroid Dehydrogenases in Late Pregnancy in Rat

Background: Aqueous extract of winter cherry (Physalis alkekengi L; family of Solanaceae) fruits (WCF) has long been recommended for fertility control by herbalists in Iran. The effect of this extract on lowering serum progesterone levels has been reported previously. Objective: To study the effects of WCF extract on the activities of ovarian 3β-hydroxysteroid dehydrogenase (3β-HSD) responsible...

Steroidogenic enzyme expression in human corpora lutea in the presence and absence of exogenous human chorionic gonadotrophin (HCG).

In a human conception cycle, the expected decline in progesterone production by the corpus luteum during the late luteal phase is prevented by human chorionic gonadotrophin (HCG) secreted by the implanting blastocyst. This study investigated the expression of components of the synthetic pathway for progesterone in human corpora lutea in the presence and absence of HCG in vivo. Corpora lutea wer...

Effects of dbcAMP on progesterone synthesis by cultured goat luteal cell subpopulations isolated from early and late luteal stage corpora lutea

This research aimed to investigate the effects of dbcAMP on steroid accumulation by culturing two distinct luteal cell subpopulations isolated from early and late luteal stage corpora lutea. Cells were isolated from corpora lutea collected from eight Angora goats on either the 5th or 15th days of their estrous cycles. Cell isolation was performed by enzymatic digestion using collagenase and DNa...

Triiodothyronine stimulates 3beta-hydroxysteroid dehydrogenase activity in the porcine corpus luteum.

OBJECTIVE To study the mechanism of thyroid hormone action on the activity of 3beta-hydroxysteroid dehydrogenase in the porcine corpus luteum. METHODS Pig ovaries were obtained from slaughterhouse animals. Luteal cells were isolated from mid-developing (5-7 days after ovulation) corpora lutea and incubated for 24 h with or without triiodothyronine. Trilostane, an inhibitor od 3beta-HSD that b...
